Amatem 20/120 Softgel is an artemisinin-based combination therapy (ACT) antimalarial medication containing 20mg of Artemether and 120mg of Lumefantrine.
Amatem combines Artemether and Lumefantrine, which have different mechanisms of action, providing a dual attack against malaria parasites.
Artemether is absorbed quickly (peak concentration at 2 hours), while Lumefantrine absorbs slower (peak at 6-8 hours). Both bind to plasma proteins. Artemether is eliminated quickly (half-life ~2 hours), while Lumefantrine has a longer half-life (3-6 days).
Dosage depends on body weight, administered over 3 days (6 doses): 15 kg to <25 kg: 12 capsules total; 25 kg to <35 kg: 18 capsules total; 35 kg and above: 24 capsules total. Specific dosing instructions are provided within the initial dose and subsequent doses over the three day period.
Contraindicated in individuals with hypersensitivity to its ingredients, those taking medications metabolized by CYP2D6, or with electrolyte imbalances. Caution during pregnancy and breastfeeding. Not evaluated for severe malaria.
Not recommended during the first trimester. Second/third trimester use only if benefits outweigh risks. Breastfeeding should be stopped during treatment and for at least one week after.
May interact with other antimalarials, CYP450 3A4 inhibitors, protease inhibitors, and antiretroviral drugs.
