How is Advant metabolized and excreted?
Advant is rapidly and completely bioactivated by ester hydrolysis during absorption from the gastrointestinal tract to candesartan. About 26% of the dose is excreted unchanged in urine. The elimination half-life of candesartan is approximately 9 hours.
Are there any special considerations for patients with renal or hepatic impairment?
Patients with severe renal impairment (creatinine clearance <30mL/min/1.73m) may experience elevated serum concentrations of candesartan. No initial dosage adjustment is necessary for patients with mild renal insufficiency.

Advant is generally well tolerated, with common side effects being mild and transient, such as headache and dizziness. Potentially important adverse events reported with Advant include asthenia, fever, paresthesia, vertigo, dyspepsia, gastroenteritis, tachycardia, palpitations, increased creatinine phosphokinase, hyperglycemia, hypertriglyceridemia, hyperuricemia, myalgia, epistaxis, anxiety, depression, somnolence, dyspnea, rash, increased sweating, and hematuria. Rare side effects include abnormal hepatic function, hepatitis, neutropenia, leukopenia, agranulocytosis, hyperkalemia, hyponatremia, renal impairment, renal failure, pruritus, and urticaria.
What should be done in case of an overdose?
The most likely manifestation of overdosage with Advant would be hypotension, dizziness, and tachycardia. Bradycardia could occur from parasympathetic stimulation. If symptomatic hypotension occurs, supportive treatment should be instituted. Candesartan cannot be removed by hemodialysis.
Are there any contraindications for using Advant?
Advant is contraindicated in patients who are hypersensitive to this drug or any component of the product. It is also contraindicated in pregnancy during the second and third trimesters.
